Describe the mechanism of P-type ATPases; select one P-type ATPase and examine how its physiological role has been investigated. The P-type ATPases are a large family of membrane enzymes, with 476 different subtypes categorised in the Swiss-Prot protein information database. The P-type or E1-E2 ATPases were first discovered and categorised by Jens Christian Skou, a Danish physician-turned-physiologist in 1957. Skou discovered the Na+/K+ ATPase, and later shared half of the 1997 Nobel Prize for Chemistry in reward for his work (Skou 1997). As mentioned above, the P-type ATPases have a broad range of interventions, for example, the use of the Digitalis toxin in the treatment of heart failure (Rang et al. 2007). This reduces the ATPases K+ affinity, causing the release of K+ into the cell (Lehninger et al. 2000). It is the above process that is responsible for approximately 25% of the bodies daily ATP breakdown and consequent energy usage (Lehninger et al. 2000). The sodium and potassium ions, and the large concentration gradients that exist, consist a vital physiological feature of the cell, and one that is implicated in myriad cellular processes. The main cellular process, as mentioned above, for which the Na/K-ATPase is responsible, is the maintenance of a membrane potential via the movements of its bonding ions. It is the maintenance, and the presence of this potential that allow the existence of the cell. The presence of a pre-existing potential allows transport of glucose and other molecules, such as Cl- ions via the mechanism of secondary active transport, harnessing the Na+ gradient into the cell caused by ATP breakdown (Skou 2004). This meant that while my friends were at dances and football games, I’d been out banishing spirits and wrestling down monsters with my stepfather. It was one of the woes of dating a veterinarian. He couldn’t help but adopt every animal he found. I couldn’t remember the cats’ names any better than the dogs’. Four of them were named after the Horsemen of the Apocalypse, and all I could really recall was that Famine ironically weighed about 30 pounds. Another problem was that Kiyo was a fox – both literally and figuratively. His mother was a kitsune, a sort of Japanese fox spirit. He’d inherited all of her traits, including amazing strength and speed, as well as the ability to transform into an actual fox. As a result, he frequently got ‘the call of the wild,’ making him yearn to run around in his animal form. Since he had downtime between jobs now, he’d left me to take a sort of wild vacation. I accepted this, but after a week of not seeing him, I was starting to get restless. â€Å"He’ll be back soon,† I said vaguely, not meeting Tim’s eyes. â€Å"Besides, you can get out of chores if you want to start paying rent.† That was our deal. I shooed him off the puzzle. Doing so took about half the puzzle pieces with him. â€Å"Goddamned cat,† I muttered. Love, I decided, was a hard thing. Well aware of my grumpy mood, I knew part of my anxiety over Kiyo stemmed from the fact that he was also passing part of his sabbatical in the Otherworld, spending time with his ex-girlfriend who just happened to be a devastatingly beautiful fairy queen. Fairies, sidhe, shining ones†¦whatever you wanted to call them, they were the tall, long-lived rulers of the Otherworld. I and most shamans referred to them as gentry, an antique term. Maiwenn, Kiyo’s ex, was almost nine months pregnant, and although they’d broken up, he was still a part of her life. I sighed. Tim might have been right about my questionable taste in men. Night wore on. I finished the puzzle while blasting Def Leppard, making me feel better. I was just shutting off the music when I heard Tim yell: â€Å"Yo, Eug. Kujo’s here.† Breathless, I ran to my bedroom door and flung it open. A red fox the size of a wolf trotted down the hall toward me. Relief burned through me, and I felt my heart soar as I let him in and watched him pace around in restless circles. â€Å"About time,† I said. He had a sleek orange-red coat and a fluffy tail tipped in white. His eyes were golden and sometimes bore a very human glint. I saw nothing like that tonight. A purely animal wariness peered out at me, and I realized it’d be a while before he changed back. He had the ability to transform to a wide range of foxes, everything from a small, normal-sized red fox to the powerful shape before me. When he spent a lot of time in this bigger form, turning human took more effort and time. Still, hoping he’d transform soon, I dumped another puzzle on my desk and worked it as I waited. Two hours later, nothing had changed. He curled up in a corner, wrapping his body in a tight ball. His eyes continued to watch me. Exhausted, I gave up on him and put on a red nightgown. Turning off the lights, I finally slipped into my bed, falling asleep instantly for a change. Introduction To The Criminal Justice System What do we mean by the Criminal Justice System (CJS)? This essay aims to answer the question What do we mean by the Criminal Justice System? In order to do this, we will start by looking at an introduction to the criminal justice system and briefing talking through the different agencies it is comprised of. We will then go on to look at the three main government departments and map out their responsibilities. These can be evaluated to determine whether or not the CJS is effective. The Criminal Justice System is one of the main public services in the UK. It is made up of specific agencies that respond to the commission of offences. These work together and share similar aims and objectives. These agencies include the Police Service, the Crown Prosecution Service, Her Majestys Court Service, The National Offender Management Service and the Youth Justice Board (cjsonline.gov.uk march 2010). All of these agencies are directed by three main government departments. These are the Home Office, the Ministry of Justice and the Office of Attorney General. These departments work together and each of these is responsible for different services within the system. The Home Office was set up in the 18th century and although many changes have been made since this time, its key objective is to protect the public therefore it is responsible for areas such as policing and security. This makes it responsible for all 43 police forces within England and Wales. It is run by the Home Secretary who sets agendas and has management boards in order to make sure its aims and objectives are carried out and their services are being delivered to the public. The Ministry of Justice was established in 2007 and took over some of the responsibilities of the Home Office. It is in charge of the justice system with areas such as criminal law and sentencing. It is now headed by the Lord Chancellor which can also be known as the Secret ary of State for Justice and like the Home Office, it also has a management board. Despite these similar goals overall, each stage of the CJS sometimes has different objectives which may result in conflict between the different agencies. For example, the courts may sentence a person to prison to be punished, while probation officers would attempt to rehabilitate them. With these differing objectives, the Office for Criminal Justice Reform (OCJR) was set up to ensure these agencies collaborated effectively and delivered the aims of the CJS. The police have the authority to refer a person to the next stage of the CJS. This is done as they have to enforce the law with the intention to keep the peace. The courts have the power to sentence an offender and a custodial sentence is the toughest punishment the courts can impose while the prisons function is to punish an offender. From this it would appear that the CJS is based upon punishment, however this is not the case. Only specific agencies focus solely on the penal system. Once an offender is sentenced by the court, the offender enters the penal system (Cavadino p1). The penal system is a system that exists to punish and otherwise deal with those who have been convicted of criminal offences (Cavadino p408). The CJS in the UK is not only about punishment but also about the rehabilitation of offenders. Chance within the play Romeo and Juliet Essay

George Aberto once said â€Å"above anything else, fate is what brings lovers together.† In Shakespeare’s play, Romeo and Juliet, fate, chance, and coincidence are all important to the development and eventually the conclusion of this tragedy. Without these three closely related themes the play would not have been able to make it out of the first act. These themes are pivotal to the development of the play and there are many instances where this is very obvious. These themes play a part in the feud between the Capulets and the Montagues, the invitation of Romeo to the ball, and the death of the two lovers, Romeo and Juliet. For as long as the two families, The Capulets and The Montagues, can remember, they have both been at war with each other and held an â€Å"ancient grudge† (Prologue). The purpose and goal of this feud is never revealed thus it must be assumed through the Prologue that their hatred is solely based on tradition from past generations. This feud is visible within the first line of Act 1 Scene 1 where Sampson, a servant of the Capulet family says that he will â€Å"not carry coals† (1.1.1). He says this in talking to another servant, Gregory, about how he will not be humiliated by a Montague servant. This allows the audience to see the importance of the feud between the Capulets and the Montagues. Romeo, a Montague, and Juliet, a Capulet, meet and fall in love at the ball in Act 1 Scene 5. This is visible in Romeos dialogue â€Å"O, she doth teach the torches to burn bright! / It seems she hangs upon the cheek of night / Like a rich jewel in an Ethiop’s ear.† (1.5.46-48). It is chance that Romeo’s true love happens to be part of the family he was brought up to hate. Romeo is aware that Juliet is a Capulet but Juliet is not aware that Romeo is a Montague and even though the feud between the families is existent, fate takes its course and allows enough time for Juliet to fall in love with Romeo. Therefore, once Juliet learns that Romeo is a Montague, she has already fallen in love with Romeo and she cannot turn back now. This is one of the most important examples of fate in the play because if she had known that Romeo was a Montague she would have never pursued a relationship with him and the rest of the play would have never turned out the way it had. Romeo and Juliet’s relationship begins at the Capulets house during the ball  without Juliet knowing that Romeo is a Montague although the fate involved in getting Romeo to the ball is just as important as Juliet not knowing Romeos last name because if Romeo would not have gone to the ball then he would have never met Juliet and fallen in love with her. It all starts with one of the Capulets servants in the streets of Verona with a list of the different guests which are invited to the ball. This list contains the names Mercutio, Tybalt, and Rosaline among others. The servant which is given the list is not able to read and this is another example of fate because once Benvolio learns that Rosaline is going to be at the ball he decides that him and Romeo will attend. Benvolio wants to â€Å"Compare her face with some that I shall show / And I will make thee think they swan a crow.† Meaning that he will bring Romeo to the ball to show that a comparison of Rosaline with other girls will make a difference and possibly relieve his sadness and love for Rosaline. This is also fate because if Rosaline was not invited, Benvolio didn’t create a plan, and if the servant would have been able to read then Romeo would have never been invited and he would have never met Juliet. The meeting of the two lovers is important although the death of the two lovers is probably one of the most important roles fate plays in the play. The first instance where fate makes its mark leading up to the death of the two lovers, Romeo and Juliet, is when the Prince banishes Romeo from the city of Verona for the death of Tybalt. The Prince reacts to the situation by saying that â€Å"for that offence / Immediately we do exile him hence.† This gets the ball rolling on the events that will lead to the two lovers death. After Romeo flees on the recommendation of Benovlio, Romeo finds himself hiding out in Friar Laurance’s Cell and then in Mantua. The next event which fate is involved in is the idea of Juliet faking her death so that she is not required to marry Paris. Friar Laurence devises a plan which he tells Juliet in Scene 1 of Act 4 as they are both talking in the Friar’s cell. He tells her to: Hold, then; go home, be merry, give consent To marry Paris: Wednesday is to-morrow: To-morrow night look that thou lie alone; Let not thy nurse lie with thee in thy chamber: Take thou this vial, being then in bed, And this distilled liquor drink thou off, (4.1. 91-96) These lines explain everything she must do so that she does not have to marry Paris and can be with Romeo instead, although he has forgotten to tell her one thing. That one thing is that the Friar has not told Juliet to tell Romeo that she is going through with this plan, faking her death. Friar Laurence realizes this and sends Friar John to give Romeo, who is hiding in Mantua, the message of his plan. What happens next is a perfect example of chance and coincidence. Right as Friar John is preparing to deliver the message an outbreak of the Plague occurs in the city of Verona and Friar John â€Å"could not send it,–here it is again,- / Nor get a messenger to bring it thee, / So fearful were they of infection† (5.2.14-16). Therefore this causes Romeo to not receive the message of the Friar’s plan but instead he receives news of Juliet’s death so it then becomes a race to the tomb, in which Juliet was placed, to stop Romeo from seeing Juliet dead. So Friar Laurence â€Å"Now must I to the monument alone; Within three hours will fair Juliet wake†(5.2.25-27) . Fates second last appearance in the play occurs Romeo arrives at the tomb first, before the Friar, and he sees Juliet, his love, â€Å"dead†. If the Friar would have arrived first, Romeo and Juliet would have never killed themselves and they would have been able to live together but that was not the case and it led to the suicide of both lovers. The final appearance of fate occurs after the lovers death and it is the end of the feud between the two families. It was fate for the lovers to die and resolve this feud. Fate, Chance, and Coincidence play an extremely pivotal part of the development of the play as well as the ending and without them the play would have turned out anything like it had and it would have never gotten  out of Act 1. The feud between the Capulets and the Montagues, Romeo’s invitation to the ball, and the death of the two â€Å"star-crossed† lovers which led to the end of the feud between the two families are all examples of how fate was able to sway the development of the story so it would lead to the death of Romeo and Juliet, â€Å"For never was a story of more woe /Than this of Juliet and her Romeo†.

The story centers on Dick Prosser, who is a black hired hand for Mr. Shepperton. Dick is involved in several levels of conflict throughout the story.* These include intrapersonal conflicts, a conflict with society, and conflict with his environment.   Ã‚  Ã‚  Ã‚  Ã‚  The first conflict is very important in the scheme of the story, because it provides the necessary conditions for this kind of an event to take place. This is Dick's conflict with his environment. Although he is a free man, and seems to be very tolerant, Dick lives in a time and place where most people are less than friendly to black men, and do not see him as an equal. This becomes evident when the auto collision occurs, and the drunk man proceeds to assault Dick without cause, and without fear of retaliation. I think I’m already a lot prepared. Whew! Wait I need to look really nice†¦ there comes my idol there! Hi Elvis! Hi Elvis!!! Good thing I have my video cam with me! Hey Elvis look at me! I’m your number 1 fan here!!! Ha-ha, nice smile over there I hope that one’s for me. I wish I was musically inclined from childhood up to now that I must have watched the concerts of my only one King of Rock and Roll! God, I never knew that he could be this famous all over the world. No one can defeat him in his place as the King. God flooded charismas all over the earth and I think he got them all. Hehe. What about his flashy hair and the prominent long side burn that every male should possess, and even a female would want to, haha. It is really his trademark in his performances, and when you ask about him they would surely give this as his first description. Anyway, who would ask about Presley when nobody does not know him? I think even people of the next generation would still remember him for his popularity and impact on the hearts of humanity. One thing I like about Elvis is how he connects to the fans, especially when he begins to sing his songs. I call him Elvis coz I know we have a connection. Many of the fans here try to imitate his hair, the way he talks and his mannerisms. Oh no two of his fans here quarrel about who looks like Elvis more, haha! Good thing about my idol is that even though he got it really a blast with the audience, he makes us feel he is just easy to reach. I remember in one of his concerts in the 70’s: â€Å"Elvis: That's The Way It Is† when he actually goes into the audience to mingle for a while–being exchanged by a lot of persons which also highlighted a party where exchanges with Elvis and Cary Grant, together with Sammy Davis Jr. appened. I never imagined that he is this down earth in joining his fans, from the most elite to the simplest type of person. That his fans only imagine this to happen but he made it all come true. Sometimes a fan collapses when he looks at them, they feel so much honored. The same with other icons like Michael Jackson, Mariah Carey and Oprah Winfrey. I also heard on his concert 7 years after â€Å" That The Way It Is† that a girl came nearer the stage to see him but unfortunately she was not able to call his attention and was partially snobbed. This girl was really desperate upon doing this, a very few thought that my idol was bad enough to neglect the fact that someone was approaching her, but then maybe he was just being nice to everyone and he wanted to treat the fans equally. One of the highlights of the life of the late Elvis was when he won an award being second all over the world after. The award given was the Golden Globe award and this was really fantastic for Elvis to have won such an honor. He also liked having a live concert because of all the electricity created on the floor. He said it was his most favorite part of the business, not the awards or whatsoever. Frank Sinatra, a friend of Elvis said that he’s going to miss him together with all the praises from the audience, his wonderful music, which only Elvis Presley can create. Even the whole America was astonished; they were really affected by this great loss, as witnessed by the former American president himself, Mr. Jimmy Carter. He showed us that the music he creates is for the benefit of all, not even just for those who know music, but even for those who will hear it first. Elvis has been influential up to now. In videoke bars the song â€Å"My Way† has always been a hit. There has been a mistake when this song is being played. They say that it has demonic influences on people listening to it and singing it. The story behind this is that everyone would like to sing the song in the microphone for this is their favorite song, and suddenly everything will go the wrong way: they will begin to shoot bad words to each other, and in the long run have some quarrel and they will resort to kill each other. Then they kill or hurt each other as if their inalienable rights were taken from them whenever the mic goes away. Everyone might have wanted to feel the electricity there is whenever Elvis Presley performed. Even so, they would like to be like him. There are still so many things about Elvis Presley that I could still remember but due to the limitations of words I could not think of ways to express them. The feeling that you want to share to other fans your personal experience about your idol would really be helpful.
